Abstract

Inthispaper,improvementinthemethodforpurifyingglycoproteinfromgreentea(Camelliasinensis)wasdescribed;somepropertiesandchemicalcompositionsofteaglycoprotein(TGP)weredeterminedbyHPGPC,FT-IR,GC–http://www.wendangwang.comparedtoexistingmethods,amorecompatiblemethodforpurifyingTGPwasproposed.Thismethodwasfaster,simpler,moree ectiveandeasiertobeextendedtotheindustrialproductionthanthemethodthatusedinourpreviouswork.ThemolecularweightofTGPwas126,513DausingHPGPC.GC–MSanalysisofTGPshowedthatTGPwascomposedofsevenkindsofmonosaccharides,namelyribose,rhamnose,arabinose,xylose,mannose,glucose,galactoseinmolarratiosof1.71:5.88:13.70:1.99:1.00:1.84:33.75.Eighteenaminoacidswereiden-ti edinTGPbyaminoacidanalysis.TheFT-IRspectrumoftheTGPrevealedalsotypicalcharacteristicsofpolysaccharides,proteinanduronicacid.

1.Introduction

Aglycoproteinisaglycoconjugateinwhichaproteincarriesoneormorecarbohydratechainscovalentlyattachedtoapolypeptidebackbone,usuallyviaN-orO-linkages(Ajitetal.,1997).Innatureglycoproteinshaveawidevarietyofbiologicalfunctions,suchasmaintenanceofproteinconformationandstability,surfaceorintracellu-larrecognition,celladhesion,processinganddiversionofprotein,andsoon.Thesefunctionsmaybedependentoneithertheproteinpartorthecarbohydratepartoronboth(Chen&Wang,1997;Sara&Jos,1997;Vliegenthart,1998).Thefastdevelopmentofbiologicalandchemicaltechnologiesduringthelast20yearshasleadtomoreandmorein-depthunderstandingonbioactivitiesoftheglycoproteins,structureofglycoproteinsandtheirrelation-ship.Itwasfoundthatglycoproteinshavevariousimpor-tantbioactivitiesinmanyaspects,suchasanti-tumor,anti-in ammation,anti-virus,loweringbloodsugar,anti-

caducity,anti-coagulant,etc.(Ajitetal.,1997;Paul,1997;Yoshida,2001).

Tea(Camelliasinensis)isoneofthe rstsetofmedicinalherbsdocumentedinancientChinesemedicinalliterature.SupposedlyoriginatedfromChina,teawasdiscoveredtobeanantidoteforpoisonousherbsbyagreatherbalist,ShenNong,about4700yearsago,whenhetestedhundredsofunknownherbsinsearchofplantswithmedicinalappli-cations.Atthesametime,lowergradegreenteawastradi-tionallyusedtocurediabeticsinEastAsia,especiallyinChinaandJapan.Todate,tea,particularlygreentea,hasbeenreportedtohaveabroadrangeoftherapeuticandnutritionalvaluesinloweringbloodsugar,bloodlipids,bloodpressure,slowingheartbeat,anti-bloodcoagulation,anti-tumor,anti-HIV,protectingbloodphase,enhancinghumannon-speci cimmunity(Marcel&Chi,2004;Rich-ard&Denis,2004;Yan,Wang,Wang,Lu,&Wang,1998).Previousresearcheshavedemonstratedthattheteaglycoconjugate,especiallyteaglycoprotein(TGP)isoneoftheactiveingredientsresponsibleformanyoftheobservedbioactivities(Chen,Zhang,&Xie,2004;Chen,Zhang,&Xie,2005;Nie,Xie,&Wang,2005;Nie,Xie,Zhou,&Cao,2006;Wang,Wang,Li,&Zhao,2001;Xie
