ABSTRACT

InordertostudywhetherbisphenolA(BPA)canpassintodrinkingwaterfrompolycarbonatebarrelandexistintheriverandindustrialef uenttheindirectcompetitiveenzyme-linkedimmunosorbentassay(ELISA)forthedeterminationofBPAwasestablished.Theresultspresentedaninhibitionconcentrationat50%absorbance(IC50)of0.123mgLÀ1,andthelimitofdetection(LOD)is9.934mgLÀ1.Thespeci cityofantiserumwasprovedwellbecausethecross-reactivitywithbenzene,tert-butylbenzene,hydroquinoneando-hydroxybenzoicacidwerefoundlowerthan0.01%,exceptphenolwas0.26%.Themethodwasfoundtobereliableandrepeatable.ItwasusedformonitoringtheconcentrationofBPAinthebarreleddrinkingwater.Theresultscon rmedBPAcanpassintobarreleddrinkingwaterfromthepolycarbonatebarrelandconcentrationincreasedasdayswenton.AcertaincontentofBPAwasfoundinindustrialef uent.TheresultsofELISAwereconsistentwiththeresultsofUV

spectrophotometry.BPAcouldnotbefoundinthewatersamplesobtainedfromZhujiangRiver.Theestablishedmethodshowsspeci crecognitionofBPAandcouldbeappliedindetectionofenvironmentalBPA.

INTRODUCTION

BisphenolAismainlyusedasaintermediateintheprepara-tionofepoxideresins,polycarbonate(PC)plasticsandasanantioxidantorstabilizerinpolyvinylchloride(Alexanderetal.1988).ExtensiveuseofBPAhasmadeitanenvironmentalcontaminantthatisfoundinsoilandriverwater(Matsumoto1982;Frommeetal.2002).Inthemeantimethewasteland llleachatesandcoatingsoncanshavealsocreatedthepossi-bilityofcontacttoBPA.TheestrogeniceffectofBPAhasbeenshownbothinvitroandinvivoexperiments(Kimetal.2001).BPAisabletomimictheeffectsofendogenoushor-mones,estrogens,andandrogens(Krishnanetal.1993),therefore,itin uencesthecentralnervoussystem(CNS)aswellasthereproductivesysteminmanyanimals(Colbornetal.1993).ExposuretolargequantitiesofBPAthroughthe

skin,however,causesextensivedamagetomultipleorgansystemsincludingthekidney,liver,spleen,pancreas,andlungs(Atkinson&Roy1955).HighdosesofBPAalsocausereproductivetoxicityandstimulategrowthoftherodentuterus(Morrisseyetal.1987).InmaleratsandmiceBPAisassociatedwithanincreasedamountofcancersofthehematopoieticsystem(Ashby&Tennant1988).Italsocausedothervariouskindsofcancers,suchasprostate,testicular,andbreastcancer,andhasdiversepleiotropicactionsinthebrainandcardiovascularsystem(Davisetal.1993;Steinmetzetal.1998).

ThestudiesmentionedaboveindicateitisimportanttodetectBPAresidues.ThemethodsmostfrequentlyusedforthedeterminationofBPAcurrentlyaregaschromatography
